Do not use if

  • You or a family member has had medullary thyroid carcinoma (MTC).
  • You have Multiple Endocrine Neoplasia syndrome type 2 (MEN 2).
  • You have had a serious allergic reaction to semaglutide or any ingredient in Rybelsus.

Common side effects

Nausea, stomach pain, diarrhea, decreased appetite, vomiting, and constipation are the most common side effects, especially when treatment is first started or the dose is increased.

Call your clinician promptly for

Persistent severe abdominal pain, ongoing vomiting or diarrhea, dehydration symptoms, vision changes, or signs of low blood sugar if you also use insulin or a sulfonylurea.

What is Rybelsus?

Rybelsus is a prescription tablet that contains semaglutide, a GLP-1 receptor agonist. It is used in adults with type 2 diabetes and is taken by mouth once daily. Serv-U Pharmacy dispenses Rybelsus only with a valid prescription.

What Rybelsus is used for

Rybelsus is indicated in adults with type 2 diabetes mellitus:

  • As an adjunct to diet and exercise to improve glycemic control
  • To reduce the risk of major adverse cardiovascular events in adults who are at high risk for these events

Your clinician decides whether Rybelsus is appropriate for your condition, current medicines, and treatment goals.

How it is taken

Take Rybelsus exactly as prescribed. It should be taken in the morning on an empty stomach with no more than 4 ounces of plain water. After taking it, wait at least 30 minutes before eating food, drinking beverages, or taking other oral medications. Swallow the tablet whole; do not split, crush, or chew.

Follow the exact strength and instructions on your prescription label. The current FDA labeling also describes a newer R2 formulation with different milligram strengths that are not substitutable on a mg-per-mg basis.

Warnings and interactions
  • Thyroid tumor warning: Rybelsus carries a boxed warning related to thyroid C-cell tumors seen in rodents; it is contraindicated in patients with a personal or family history of MTC or with MEN 2.
  • Pancreatitis: Stop the medicine and seek medical advice right away for persistent severe abdominal pain, with or without vomiting.
  • Diabetic retinopathy: Patients with a history of diabetic retinopathy should be monitored.
  • Hypoglycemia risk: Risk may be higher if used with insulin or sulfonylureas.
  • Kidney problems from dehydration: Ongoing nausea, vomiting, or diarrhea can lead to dehydration and kidney injury.
  • Severe GI problems: Rybelsus is not recommended in patients with severe gastroparesis.
  • Other oral drugs: Because Rybelsus delays gastric emptying, your clinician or pharmacist may need to review the timing of other oral medicines.
Missed dose guidance

If you miss a dose, skip the missed dose and take the next dose the following day. Do not take two doses on the same day.

Store at room temperature in the original container, protected from moisture, and keep out of reach of children.

Patients often take the tablet with coffee, breakfast, or other morning medications, and that can reduce absorption. The best routine is simple: keep the bottle next to your bed, take one tablet as soon as you wake up with a small amount of plain water only, then wait at least 30 minutes before food, coffee, supplements, or other pills. Another common issue is dose-escalation nausea. That is why it helps to confirm your next strength before you finish your current bottle and to stay well hydrated if stomach upset occurs. If you also use insulin or a sulfonylurea, pay attention to low-blood-sugar symptoms and ask your clinician whether any dose adjustments are needed after starting semaglutide therapy.

Can I take Rybelsus with coffee or breakfast?
No. Take it first with plain water only, then wait at least 30 minutes before food, coffee, or other beverages.

Can I take my other morning pills at the same time?
No. Wait at least 30 minutes after taking Rybelsus before taking other oral medications unless your prescriber tells you otherwise.

What if I miss a dose?
Skip the missed dose and take your next dose the following day. Do not double up.
